Pancreatic fat cells of humans with type 2 diabetes display reduced adipogenic and lipolytic activity

چکیده

Obesity, especially visceral fat accumulation, increases the risk of type 2 diabetes (T2D). The purpose this study was to investigate impact T2D on pancreatic depot. Pancreatic pads from 17 partial pancreatectomized patients (PPP) were collected, preadipocytes isolated, and in vitro differentiated. Patients grouped using HbA1c into normal glucose tolerant (NGT), prediabetic (PD), T2D. Transcriptome profiles adipocytes assessed by RNAseq. Insulin sensitivity estimated quantifying AKT phosphorylation Western blots. Lipogenic capacity with oil red O staining, lipolytic activity via fatty acid release. Secreted factors measured ELISA. Comparative transcriptome analysis indicates defective upregulation genes governing adipogenesis ( NR1H3), lipogenesis FASN, SCD, ELOVL6, FADS1), lipolysis LIPE) during differentiation cells T2D–PPP. In addition, ratio leptin/adiponectin mRNA higher than NGT–PPP. Preadipocytes NGT–PPP more insulin sensitive T2D–PPP regard phosphorylation. Triglyceride accumulation similar NGT adipocytes. Despite a high expression receptors NPR1 NPR2 adipocytes, stimulated ANP 1.74-fold only. This stimulation further increased PDE5 inhibitor dipyridamole (3.09-fold). Dipyridamole forskolin receptor independently 1.88-fold 1.48-fold, respectively, solely cells. conclusion, metabolic status persistently affects These alterations could aggravate development

Reduced Insulin Exocytosis in Human Pancreatic β-Cells With Gene Variants Linked to Type 2 Diabetes

The majority of genetic risk variants for type 2 diabetes (T2D) affect insulin secretion, but the mechanisms through which they influence pancreatic islet function remain largely unknown. We functionally characterized human islets to determine secretory, biophysical, and ultrastructural features in relation to genetic risk profiles in diabetic and nondiabetic donors. Islets from donors with T2D...

Association of Physical Activity level with the COVID-19 Incidence in Patients with Type 2 Diabetes

Background and Objective: Type 2 Diabetes Mellitus (T2DM) is among the epidemic metabolic chronic diseases that attenuates physiological functions, including hematopoiesis and immune responses, developing a higher risk of COVID-19 incidence. This study purposed to study the association of physical activity level (PAL) with the COVID-19 incidence in patients with T2DM. Materials and Methods: Al...

association of physical activity with risk of type 2 diabetes

background: physical activity has shown to prevent type diabetes 2. however, the type, intensity and amount of effective physical activity as well as individuals' needs according to level of their risk for type 2 diabetes have not been clarified comprehensively. this study investigated a relation between moderate aerobic physical activity ≥150 minuets/week with decreased risk of type 2 diabe...

Adipose Mesenchymal Stromal Cells Isolated From Type 2 Diabetic Patients Display Reduced Fibrinolytic Activity

Stem cells have been successfully used for the treatment of critical limb ischemia (CLI). We conducted a clinical trial to determine the feasibility of using autologous adipose-derived mesenchymal stromal cells (AdMSCs) for the treatment of CLI. Unexpectedly, two diabetic patients developed peripheral microthrombosis. This adverse effect, which contrasts with the reported antithrombotic propert...
